Guess I'll have to weigh in with the opinion that it comes from the military "by
the numbers." For the command hand salute, one is bring your right hand to the
eyebrow in a salute. Ready, two, is drop the hand back to your side.
The "two" command is given by the person in charge to signify the end of that
step of the activity.
For those commands with multiple steps, you might have the initial command plus
ready, two; ready, three; ready, four, etc.
